Shellfish collected as part of the UK biotoxin statutory monitoring programme (SMP) are currently analysed for PSP toxins using the AOAC mouse bioassay (MBA). Much effort has however been applied to reduce reliance on such animal testing. The AOAC HPLC method validated by Lawrence and drafted in the European legislation as an alternative to the bioassay is being trialled in Europe. A recent inter-laboratory trial however highlighted a number of issues with the method, which may delay its implementation in national SMP.

As an alternative, Cefas and the Food Standards Agency, the UK Competent Authority, evaluated the performance of a variation of the Lawrence method, which would allow the screening, by HPLC, of shellfish for PSP toxins. Only those samples found HPLC positive would be forwarded to MBA for determination of toxin content. The method was successfully validated and parallel tested with the MBA to demonstrate fitness for purpose and robustness. Methodology and results will be presented to show that the method is suitable for the screening of mussels, oysters, king scallops and cockles. This method will be implemented from 1st November 2006 in the GB biotoxin SMP, achieving an expected reduction in animal usage for PSP testing of ~70% pa.
